Nicotinamide affects skin pigmentation by reducing the movement of melanosomes from melanocytes to keratinocytes (without inhibiting tyrosinase activity or cell proliferation). It also reduces melanin production by interfering with the cell signaling pathway between keratinocytes and melanocytes. Furthermore, nicotinamide acts on existing melanin, reducing its movement to skin surface cells.

Niacinamide promotes protein synthesis in the epidermis and skin cell regeneration, helping to shed aged skin cells and form a healthier skin cell structure. This promotes the rapid generation of new skin cells.

Additionally, the amine group in nicotinamide neutralizes free fatty acids in sebum, reducing their concentration within pores. This minimizes skin inflammation and reduces the risk of acne, resulting in smoother, clearer skin.
